Abstract

[Abstract] Objective: To establish a liquid chromatography-triple quadrupole mass spectrometry (LC-MS / MS) method for the determination of sialic acid N-acetylneuraminic acid (Neu5Ac) and N-hydroxyacetylneuraminic acid (Neu5Gc) in visceral and muscle tissues of pigs. And Deaminine Neuraminic Acid (KDN) Concentration Analysis Method. Methods: After homogenizing the visceral meat tissue, trifluoroacetic acid (TFA) was used to dissociate the sialic acid from the glycolipid or glycoprotein chain and then analyze the sample. Chromatographic conditions: ZORBAX Eclipse Plus C18 (5 μm, 4.6 × 250 mm, Waters), mobile phase: 0.1% ammonium acetate-100% acetonitrile (9: 1), column temperature: 25 ° C, flow rate 800 μL · min -1 The injection volume was 20 μL. Mass spectrometry conditions: the ion source is electrospray ionization source (ESI), the scanning method is multiple reaction monitoring (MRM), and the monitoring ion pairs: Neu5Ac: 307.7 → 87.0, Neu5Gc: 323.3 → 116.0, KDN: 266.7 → 87.0, 13C3Neu5Ac internal standard Product: 310.9 → 90.0.
