The urinary steroid profile of geldings with 'elevated' endogenous production compared to those administered anabolic agents (pending)

Abstract

Detection of the use of testosterone or testosterone precursors in geldings is regulated by an international threshold of 20 ng/ml free and conjugated testosterone in urine and 100 pg/ml free testosterone in plasma. Furthermore, detection of an intact steroid ester in blood and/or urine provides direct evidence of intentional administrationAs previously presented at ICRAV 2012, there has been an apparent increase in the incidence of 'elevated' testosterone in gelding urine samples tested by Racing NSW in Australia since around 2010. More recently, a similar phenomenon has been observed in gelding urine samples tested in the UK. However, the UK samples were also often associated with elevated hydrocortisone concentrations. Whilst a small number of the affected UK samples breached the international urinary testosterone threshold, the overall steroid profiles were not consistent with those expected following the administration of testosterone or a testosterone precursor.To coordinate approaches on these apparently similar issues, the Australian Racing Board (ARB)/Racing NSW and BHA/LGC have collaborated in a number of areas. This presentation will provide an overview of this collaboration as well summarising the results of those studies conducted in the UK.Parallel strands of research/surveillance included population studies, longitudinal testing of a number of the affected animals as well as investigations into the effects of a number of drug administrations on the plasma and urine steroid profiles. Following these investigations, a range of statistically modelled urinary steroid ratios were found to be capable of differentiating elevated endogenous production of testosterone from the administration of testosterone esters, Synacthen (synthetic ACTH), porcine growth hormone, DHEA (administration conducted by Charles Sturt University/ARB/Racing NSW) and the product Equibolic (a preparation purported to contain a 1:1 mixture of pregnenolone and DHEA).
